A Study on the Culture of Pluteus cervinus


Abstract
Cultural characteristics of Pluteus cervinus ISA-Pe-1004, which was newly recognized as an antitumor protein-polysaccharide producing basidiomycete, were studied in shake-culture flasks.
This fungus reached full growth in 7 days when cultured at 28¡Æ, 180 rpm. The optimal pH was 4.5 and good growth was observed in the pH range from 4.0 to 5.5. Yeast extract was found to be the most effective nitrogen source when used alone or together with peptone. The optimal inoculum size was in the range of 5¡­10% (v/v) when 7-day old culture was used as inoculum. This fungus showed good growth in high consistency media of the total concentration up to 20%. Thirty-seven grams of dried mycelia were obtained per liter of medium when cultured in high consistency media (12%) at 28¡Æ for 7 days. The composition of this medium was glucose 85.8g, yeast extract 17.lg, peptone 17.1g, KH©üPO©þ 0.87g, MgSO©þ¡¤7H©üO 0.5g, CaCl©ü 0.3g, FeSO©þ¡¤7H©üO 1§·, ZnSO©þ¡¤7H©üO 4§·, CuSO©þ¡¤5H©üO 1§·, MnCl©ü¡¤4H©üO 7§·, per liter.
